Indirect Shoot Regeneration Using 2,4-D Induces Somaclonal Variations in Cinchona officinalis

Cinchona officinalis is an important species from the Andean cloud forest that has a low regeneration rate in natural populations. In vitro of C. been successfully established but somaclonal variation was not evaluated. The pathway and number subcultures on were evaluated using six ISSR primers amplified 58 loci Inter Simple Sequence Repeats (ISSR). A dendrogram based Jaccard´s genetic distance between donor plant produced. results show indirect shoot induces variation, presence 2,4-Dichlorophenoxyacetic acid (2,4-D) combination with kinetin 6-Benzylaminopurine (BAP). 1-Naphthaleneacetic (NAA) or Indole-3-butyric (IBA), BAP produces genetically stable explants. highest proliferation achieved IBA. present research study suggests avoiding use 2,4-D when propagated for reintroduction restoration projects.
